Molecular process in fat cells that influences stress and longevity identified

ScienceDaily (Sep. 26, 2012) ? As part of their ongoing research investigating the biology of aging, the greatest risk factor for type 2 diabetes and other serious diseases, scientists at Joslin Diabetes Center have identified a new factor -- microRNA processing in fat tissue -- which plays a major role in aging and stress resistance. This finding may lead to the development of treatments that increase stress resistance and longevity and improve metabolism.

The findings appear in the Sept. 5 online edition of Cell Metabolism.

Over the past several years, it has become clear that fat cells (adipocytes) are more than just repositories to store fat. Indeed, fat cells secrete a number of substances that actively influence metabolism and systemic inflammation. Previous studies have found that reducing fat mass by caloric restriction (CR) or surgical or genetic means can promote longevity and stress resistance in species from yeast to primates. However, little is known about how CR and fat reduction produce these beneficial effects. This study investigated one type of molecular mediator -- change in microRNAs (miRNAs) and the processing enzymes required to make them- that is influenced by aging and reversed by caloric restriction. miRNAs are involved in the formation of mature RNA.

Based on studies conducted using human cells, mice and C. elegans (a microscopic worm used as a model organism for aging studies), the researchers demonstrated that levels of multiple miRNAs, decrease in fat tissue (adipose) with age in all three species. This is due to a decrease in the critical enzyme required from converted pre-miRNAs to mature miRNAs, Dicer. In the human study, which compared the miRNA levels in preadipocytes (fat cell precusors) of young, middle-aged and older people, people aged 70 and older had the lowest miRNA levels. "The fact that this change occurs in humans, mice and worms points to its significance as a general and important process," says lead author C. Ronald Kahn, MD, Chief Academic Officer at Joslin Diabetes Center and the Mary K. Iacocca Professor of Medicine at Harvard Medical School.

Caloric restriction, which has been shown to prolong lifespan and improve stress resistance in both mice and worms, prevents this decline of Dicer, and in the case of the mice, restore miRNAs to levels observed in young mice. Conversely, exposure of adipocytes to major stressors associated with aging and metabolic diseases, including toxic agents, Dicer levels decreased. Mice and worms engineered to have decreased Dicer expression in fat showed increased sensitivity to stress, a sign of premature aging. By contrast, worms engineered to "overexpress" Dicer in the intestine (the adipose tissue equivalent in worms) had greater stress resistance and lived longer.

Overall, these studies showed that regulation of miRNA processing in adipose-related tissues plays an important role in longevity and an organism's ability to respond to age-related and environmental stress. "This study points to a completely new mechanism by which fat might affect lifespan and is the first time that anyone has looked at fat and miRNAs as factors in longevity," according to co-author T. Keith Blackwell, MD, PhD, co-head of Joslin's Section on Islet Cell and Regenerative Biology and Professor of Pathology at Harvard Medical School.

Based on this study, Blackwell suggests that "finding ways to improve miRNA processing to keep miRNA levels up during aging might have a role in protecting against the stresses of everyday life and the development of age- and stress-related disease."

Dr. Kahn and the study investigators are currently working on ways to genetically control Dicer levels in the fat tissues of mice, to create mouse models that are more or less resistant to stress. "We would love to find drugs that would mimic this genetic manipulation to produce a beneficial effect," says Dr. Kahn. "If we can better understand the biology of aging, we might also understand how age impacts diabetes," says Kahn.

Study co-authors include Marcelo A. Mori, Prashant Raghavan, Jeremie Boucher, Stacey Robida-Stubbs, Yazmin Macotela, Steven J. Russell, and T. Keith Blackwell of Joslin; and James L. Kirkland and Thomas Thomou of the Mayo Clinic.

Would Hemingway Blog? ? Kristen Lamb's Blog

Photo by Yousef Karsh via Wikimedia Commons

Emphatically, YES!!! I know many writers are hesitant to the idea of blogging. It feels like just another social media chore, but nothing can be farther from the truth. In fact, blogging is probably the ONLY form of social media that 1) draws from a writer?s strengths and 2) doesn?t try to fundamentally change our personality.

Yes, as a social media expert Jedi, I will tell you that it?s a good idea to tweet and learn to use Facebook, but I?m also going to tell you something you already know. Most of that kind of social media is NOT natural for a lot of writers. Is it good for you? Yes. It shoves you out of your comfort zone and makes you work an area that will be vital to career success. But, of all the various on-line tools we can wield, the blog is by far one of the best.

Oh, but Kristen. There are already way too many blogs out there.

Yep, and guess what? There are way too many books out there, too, and that hasn?t stopped you guys from writing one, has it? Blogs are a lot like books. In fact, that is one of the reasons they are such an excellent choice for writers. Blogs connect using?words. Same as books. They connect through information or emotion?same as books. If people learn to love your blogging voice, it is no great leap to love your novels.

Ah, but just like books?

Most people who start a book never keep pressing until it is finished. Similarly, most people who start a blog will abandon it for some new shiny two months in. Most people who start writing a novel believe it is easy, and that they don?t need any professional instruction or guidance. Guess what? Same with blogs.

Too many people who start a blog just throw up content without learning what to blog, how to blog, and what makes a blog grow and become successful. This means the competition is not nearly as daunting as some might believe.

So why would Hemingway blog? Well, actually, he did. I am going to paraphrase a story relayed by mega-author and Hemingway expert, David Morrell.

Hemingway was a Blogger Journalist

As a young reporter for the Kansas City Star, Hemingway learned the value of lean, uncluttered sentences. In fact, the newspaper?s style sheet underscored the, ?Use of vigorous English?Be positive?Avoid the use of adjectives.? Though Hemingway followed these principles as a reporter, he apparently forgot them when he decided to write fiction. When he moved to Paris and showed Getrude Stein his work, she slayed him for his purple prose. She told him to toss everything and try again.

A few months later, Hemingway met a reporter in Switzerland who expressed interest in his work. Hemingway was so excited he wrote to his wife and asked her bring all of his manuscripts to him straight away. Being a good wife, she packed everything he?d written in a suitcase and hopped on a train?and the suitcase was stolen, taking every shred of Hemingway?s writing.

Hemingway rushed home and turned his apartment upside down, but to no avail. It was all gone. Hemingway almost gave up, but then he thought back to Gertrude Stein?s advice to chuck everything and begin anew. Hemingway rolled up his sleeves and went back to work, yet this time he harnessed his reporting skills and went about his writing in a far more organized fashion, with the verbal discipline he?d learned from the Kansas City Star.

Hemingway learned that less is more, that economy of description can produce clearer effects than descriptions with detail piled upon detail. But, economy doesn?t only mean reducing a description to its essentials. It also means going for so clean a line that adjectives and adverbs become a sign of bad writing. ~David Morrell The Successful Novelist p.117

I have been running my writing contest for over two years now, and I see the same problems over and over with new writers. The prose is bogged down with all kinds of fluff. The sentences aren?t clear and the prose is weak.

Just like Hemingway used his experience as a reporter to strengthen his fiction (which made him one of the greatest writers in literary history), we, too, can use blogging to refine our prose and strengthen our writing skills. There are many great authors who used their journalistic muscles to write great works of fiction. Hemingway, Orwell, Dickens and Twain to name a few.

Blogging is a modern equivalent of journalism, and I believe Hemingway definitely would have blogged had he been a man of a different era. Can you imagine Hemingway tweeting images of giant swordfish he?d caught deep sea fishing? Or posting a video on You Tube of him running with the bulls? Maybe some images on Flikr from his latest safari?

Where was I? Oh, yes!

Blogging Takes Us from Neophyte to Expert MUCH Quicker

Malcolm Gladwell asserted that it takes 10,000 hours of practice to become an expert. Add blogging to your repertoire, and guess how quickly you can rack up that 10,000 hours? Blogging will teach you to write faster, leaner and with far more power in much less TIME.

Bloggers Learn to Ship

When you have a blog due, you learn to kill Little Darlings with ruthless efficiency. Bloggers (like journalists) learn not to grow overly fond with sections of prose. We are copy editing MACHINES. We are great at meeting deadlines because we don?t need 42 different opinions to convince us to part with some prose.

HACK! HACK! SLICE!

Bloggers Grow a Thick Skin

Writers who also blog are showing the world they take their profession seriously. We put our work out there, good, bad or WTH?. We open ourselves to criticism, and we learn to take it like a champ and come back swinging.

I?ve met a lot of writers who get defensive, angry or abusive when told their work isn?t a glittery kitten hug. This business is tough, and blogging will whip a writer into fighting form in no time.

Blogging Trains Us for Other Paid Work

Since blogging is so close to journalism, it is easier for us to get paid work writing articles, blogs or even copy work. Bloggers have a BLOG that shows the world that they are serious. Potential employers see a writer who can make deadlines, who can work even when they don?t feel like it. Bloggers, like journalists, don?t sit around and wait for the inspiration fairy. They roll up their sleeves and do what real writers do.

They write.

Additionally, many writers supplement their book income with other work (like articles), and blogging is a great way to get your foot in the door.

Getting Started

So for those who don?t want to blog, that is fine. But for those who do?

Blogging is one of the best ways to build an author platform (mainly because it has us operating in our strength?writing). A blog is far less volatile than other forms of social media. Who knows if we will have Twitter in five years? Twitter may go, but a blog will remain and can continue to grow for YEARS. We don?t have to be a Chatty Cathy social butterfly to be a kick@$$ blogger, and this is really great for those shy introverts out there. In fact, in my experience, you guys make some of the BEST bloggers.

Starting a Successful Blog

A lot of blogs fail simply because writers take off with no instruction, and, because of this, they are left to learn by painful trial and error. If you believe you would like to blog, but you?re uncertain, I?m doing something new. To accommodate those who are still on the fence, I?m now running a Basic level for my upcoming blogging class.

In the Basic class, you get to be part of the WANA1012 team and receive all the forum lessons (none of the live webinars are included). This is a really great place to learn if blogging is right for you (Blogging Training Wheels).

If you?re ready to skip the training wheels and get started blogging, then get your spot NOW. My classes have a history of selling out. I offer a Blogging Bronze, Silver, Gold, and even Diamond, for those who are ready to go all the way.

This is a TWO MONTH class?one month for lessons and one for launch?that you can do in your own time, at your own speed and from home. And since you will be part of a WANA team, you won?t have to do this blogging thing alone, so your odds of success are MUCH higher. For those who want to do NaNoWriMo, we can extend the two months if we have to. That?s one of the benefits of being the owner of the interface :D .

Anyway, what are your thoughts? Those of you who blog, have you seen an improvement in your writing? What questions do you guys have? Thoughts? What other famous writers from history would be cool to see tweeting or posting blogs? Poe? Steinbeck? Shakespeare? What do you think would be their favorite social site and how would they use it? Picasso and Pinterest? :D

I LOVE hearing from you guys!

Oh, and if you love this blog, I would love your support. I am in the running to become a community blogger for my hometown, so I?d appreciate your votes. Just click the link and scroll down until you see my name and vote. THANK YOU! When the zombie apocalypse arrives, I promise to share ammo and Twinkies with those of you who vote for me :D .

Back to the regular contest?.

To prove it and show my love, for the month of September,?everyone who leaves a comment I will put your name in a hat. If you comment and link back to my blog on your blog, you get your name in the hat twice. If you leave a comment, and link back to my blog, and mention my book?We Are Not Alone?in your blog?you get your name in the hat THREE times.?What do you win? The unvarnished truth from yours truly.

I will pick a winner?once a month?and it will be a critique of?the first 20 pages of your novel,?or your query letter, or your synopsis (5 pages or less).

And also, winners have a limited time to claim the prize, because what?s happening is there are actually quite a few people who never claim the critique, so I never know if the spam folder ate it or to look for it and then people miss out. I will also give my corporate e-mail to insure we connect and I will only have a week to return the 20 page edit.

At the end of September I will pick a winner for the monthly prize. Good luck!

I also?hope you pick up copies of my best-selling books? We Are Not Alone? The Writer?s Guide to Social Media?and? Are You There, Blog? It?s Me, Writer?.?And both are recommended by the hottest agents and biggest authors in the biz. My methods teach you how to make building your author platform FUN. Build a platform and still have time left to write great books.

Mechanism that leads to sporadic Parkinson's disease identified

ScienceDaily (Sep. 25, 2012) ? Researchers in the Taub Institute at Columbia University Medical Center (CUMC) have identified a mechanism that appears to underlie the common sporadic (non-familial) form of Parkinson's disease, the progressive movement disorder. The discovery highlights potential new therapeutic targets for Parkinson's and could lead to a blood test for the disease. The study, based mainly on analysis of human brain tissue, was published September 25 in the online edition of Nature Communications.

Studies of rare, familial (heritable) forms of Parkinson's show that a protein called alpha-synuclein plays a role in the development of the disease. People who have extra copies of the alpha-synuclein gene produce excess alpha-synuclein protein, which can damage neurons. The effect is most pronounced in dopamine neurons, a population of brain cells in the substantia nigra that plays a key role in controlling normal movement and is lost in Parkinson's. Another key feature of Parkinson's is the presence of excess alpha-synuclein aggregates in the brain.

As the vast majority of patients with Parkinson's do not carry rare familial mutations, a key question has been why these individuals with common sporadic Parkinson's nonetheless acquire excess alpha-synuclein protein and lose critical dopamine neurons, leading to the disease.

Using a variety of techniques, including gene-expression analysis and gene-network mapping, the CUMC researchers discovered how common forms of alpha-synuclein contribute to sporadic Parkinson's. "It turns out multiple different alpha-synuclein transcript forms are generated during the initial step in making the disease protein; our study implicates the longer transcript forms as the major culprits," said study leader Asa Abeliovich, MD, PhD, associate professor of pathology and neurology at CUMC. "Some very common genetic variants in the alpha-synuclein gene, present in many people, are known to impact the likelihood that an individual will suffer from sporadic Parkinson's. In our study, we show that people with 'bad' variants of the gene make more of the elongated alpha-synuclein transcript forms. This ultimately means that more of the disease protein is made and may accumulate in the brain."

"An unusual aspect of our study is that it is based largely on detailed analysis of actual patient tissue, rather than solely on animal models," said Dr. Abeliovich. "In fact, the longer forms of alpha-synuclein are human-specific, as are the disease-associated genetic variants. Animal models don't really get Parkinson's, which underscores the importance of including the analysis of human brain tissue."

"Furthermore, we found that exposure to toxins associated with Parkinson's can increase the abundance of this longer transcript form of alpha-synuclein. Thus, this mechanism may represent a common pathway by which environmental and genetic factors impact the disease," said Dr. Abeliovich.

The findings suggest that drugs that reduce the accumulation of elongated alpha-synuclein transcripts in the brain might have therapeutic value in the treatment of Parkinson's. The CUMC team is currently searching for drug candidates and has identified several possibilities.

The study also found elevated levels of the alpha-synuclein elongated transcripts in the blood of a group of patients with sporadic Parkinson's, compared with unaffected controls. This would suggest that a test for alpha-synuclein may serve as a biomarker for the disease. "There is a tremendous need for a biomarker for Parkinson's, which now can be diagnosed only on the basis of clinical symptoms. The finding is particularly intriguing, but needs to be validated in additional patient groups," said Dr. Abeliovich. A biomarker could also speed clinical trials by giving researchers a more timely measure of a drug's effectiveness.

The study was supported by the grants from the Michael J. Fox Foundation, the National Institutes of Health, and the National Institute of Neurological Disorders and Stroke (RO1NS064433).

(BN) Xbox Challenged as Cable Plots to Make Consoles Obsolete

AT&T Inc. (T), Verizon Communications Inc. (VZ) and Time Warner Cable Inc. (TWC) are gearing up for a push to deliver video games directly to televisions, said people with knowledge of the matter, a strategy shift that poses a threat to traditional consoles such as the PlayStation, Wii and Xbox.

Trials of cloud-gaming services are likely to start later this year so carriers can test and tweak the technology before wider deployments that may begin as early as 2013, said the people, who asked not to be named because the discussions are private. Other carriers are aiming for 2014, the people said.

If successful, Web-based games could accelerate a shift away from consoles, the industry's main money maker for the past three decades. Sony Corp. (6758), Microsoft Corp. (MSFT) and Nintendo Co. have helped to build a market worth $24.1 billion in the U.S. in 2011, according to NPD Group Inc. Consumers are already dumping consoles in favor of games on smartphones and tablets, leading to a 39 percent decline in video-game hardware sales last month from a year earlier.

"Everybody has a TV," said Atul Bagga, a video-games analyst at Lazard Capital Markets in San Francisco. Cable and phone companies are "looking for new ways to monetize their users and gaming can be pretty compelling," he said.

By adding popular games to their TV, Internet and phone packages, carriers can offer another service to their almost 50 million digital TV subscribers.

Real Time

In addition to AT&T, Verizon and Time Warner Cable, Comcast Corp. (CMCSA) and Cox Communications Inc. are also in talks to offer video-gaming services, the people said. They're all looking to go beyond social games from Zynga Inc. (ZNGA) and casual games such as "Tetris" and "Solitaire," with technology that can deliver the most advanced action games from top publishers such as Electronic Arts Inc. (EA)

For technology, the carriers are turning to startups such as Playcast Media Systems, CiiNOW Inc. and Agawi Inc., which provide software to speed delivery of real-time gaming. Executives at each of those companies acknowledged that they're in talks with U.S. carriers, declining to say which ones.

Alex Dudley, a spokesman for Time Warner Cable, which had 12.3 million subscribers as of mid-2012, and Jennifer Khoury, a spokeswoman for Comcast, which had 22.1 million subscribers, declined to comment.

Jan Rasmussen, a spokeswoman for AT&T, said in a statement the company is "exploring unique ways to offer cloud gaming services to our TV and broadband customers." AT&T had 4.15 million subscribers for its U-verse TV services as of June.

Console Model

Deidre Hart, a spokeswoman for Verizon, said that while the company has the capability, it doesn't currently "offer anything regarding HD cloud gaming." Shana Keith, a spokeswoman for Cox, said the company is exploring a number of cloud-based broadband services, declining to provide specifics.

Verizon had 4.47 million TV subscribers, while Cox had 4.66 million, according to the National Cable & Telecommunications Association.

With cloud gaming, consumers will be able to avoid buying Sony's PlayStation 3, Microsoft's Xbox 360 or Nintendo's Wii, and play using generic controllers connected to their set-top box or TV. Some carriers are looking at software that turns smartphones into controllers, the people said.

Wider cloud-gaming adoption would be a significant shift from the console business model pioneered by Atari Inc. in the '70s and advanced by Nintendo in the '80s -- selling machines at a slim profit or loss and making money on royalties from game cartridges or discs.

Jigsaw Puzzle

Manufacturers have built online accounts and data centers for current console subscribers in order to offer games, interactive environments and prepare for networked gaming. Xbox Live, Microsoft's online service, lets gamers play together, buy games and use other devices.

Carriers still have to get the technology in place. To stream games from remote servers to multiple devices simultaneously, they need to license virtualization technology. And to make the experience comparable to that of a console, they also must incorporate powerful graphics processors into their data centers, replacing chips used in consoles.

Putting all those pieces together proved too difficult for OnLive Inc., a startup backed by units of AT&T and Time Warner Inc., which went through a restructuring last month after failing to attract enough $9.99-a-month subscribers to its cloud-gaming system. Gaikai Inc., a competitor in the market, agreed to be bought earlier this year by Sony for $380 million.

Execution Challenge

Large service providers have an advantage because they have deeper pockets, big data centers and an existing subscriber base. Still, delivering a cloud service with the same quality as game consoles and creating a profitable business will be a challenge, said Mitch Lasky, a partner at venture firm Benchmark Capital in Menlo Park, California, and an early investor in Gaikai.

"It makes perfect sense why they would want to go after this market," said Lasky, who was previously an executive at Electronic Arts. "Streaming games use a ton of bandwidth and really benefit from good networks. But it's a gnarly execution problem they're trying to solve."

Nvidia Corp., a maker of graphics processors, has been building technology for data-centers and said earlier this year that it's working on implementing it in a way that game service providers can use. The company is making a big bet on cloud gaming, said Tony Tamasi, a senior vice president.

"It's a substantial investment of both hardware and software," said Tamasi, though he declined to name the company's partners. "We've put stuff into our chips specifically to enable this kind of functionality."

Stream Games

For game publishers, cloud gaming makes sense because they can develop for a single platform rather than for each of the various consoles, which costs more money. Frank Gibeau, president of Electronic Arts Labels, said games via Web-based TV will eventually be a "big opportunity," without giving a time frame.

While networks in the U.S. are preparing for cloud gaming, services have already emerged in other countries, including Portugal, France, Singapore and South Korea. Playcast, based in Israel, has partnered with carriers in each of those countries to stream games to set-top boxes and televisions.

In July, Playcast announced a deal with South Korea's CJ Hellovision to make a gaming service available to the cable operator's 3.51 million subscribers. Last year, Playcast partnered with Singapore Telecommunications Ltd. (ST) for a S$9.99 ($8) a month subscription service that included titles from Activision Blizzard Inc. (ATVI) and Atari.

CiiNOW, based in Mountain View, California, is about to begin its fourth European trial, though Chief Executive Officer Ron Haberman declined to name the carriers. U.S. cable and phone companies need to catch up, he said.

"If there was ever a service that fit network providers, it's this one," Haberman said. "2013 is going to be when we see big commercial offerings."
